Product: Recombinant streptolysin O. Functional in membrane pore   formation to introduce molecules into living animal cells. The product was highly purified from E.coli over-expressing SLO of Group C hemolytic streptococci (His6-tagged to the signal peptide removed N-terminal of SLO). The specific activity is as high as 1,200,000- 2,000,000 hemolytic units (HU) /mg (depending on Lot).

Measurement of the activity: Definition of 1HU is activation of 50% hemolysis by incubating 3% sheep red blood cells at 37℃ for 30 min.

Streptolysin O (SLO) is a membrane-damaging extracellular toxin produced by hemolytic streptococci. In the hemolytic streptococci infection the antibodies to various proteins secreted from the bacteria such as toxins increase. For the diagnostic confirmation of SLO production by the infection the measurement of the antibody ASLO (Anti-Streptolysin O) is most widely used.

NAD (nicotinamide adenine dinucleotide) hydrolyzing enzyme is one of the extracellular enzymes and toxins produced by hemolytic streptococci. Although its function as a toxin is largely unknown it has been suggested to be related to pathogenicity of acute infection (1). NADase is produced not only by Group A hemolytic streptococci but also by Group C and Group G strains. The amino acid sequences are highly conserved among them and the antibodies cross-react each other. Upon infection of hemolytic streptococci the antibody titer to the NADase increases similarly to anti-SLO (Strreptolysin O) antibody.

ATF6 (activating transcription factor 6) is an endoplasmic reticulum (ER) membrane-bound transcription factor activated in response to ER stress. When unfolded proteins accumulate in the ER ATF6 alfa is cleaved by regulated intramembrane proteolysis. The resulting amino-terminal fragment translocates to the nucleus and activates transcription by binding to ER stress-response elements present in the promoter regions of ER stress-inducible genes including those encoding ER chaperones and components of ER-associated degradation. ATF6 consists of two closely related factors ATF6 alfa and ATF6 beta in mammals. ATF6 alfa but not ATF6 beta plays a pivotal role in transcriptional control.
The monoclonal antibody was characterized in the laboratory of Professor Kazutoshi Mori of Kyoto University. The antibody was produced from hybridoma cultured in serum-free medium and purified under mild conditions by propriety chromatography processes.
