E. coli LexA protein binds specifically to the SOS-box sequence and represses the genes belonging to the SOS regulon. In response to DNA damage RecA protein is activated by ss-DNA accumulated in the damaged cells and promotes autocleavage of LexA repressor by its coprotease activity. As a result DNA repair genes and error prone polymerases are induced and DNA damage is repaired and mutation is induced (1).
The lexA gene is used for yeast two-hybrid experiments as a bait to identify the protein-protein interaction in vivo (2).
This product was prepared by immunizing rabbit with full-size highly-purified recombinant LexA protein. Using this antibody 23 kD LexA protein was identified in the E. coli whole-cell lysate (Fig 1) and the expression of bait constructs was identified in yeast extracts by Western blotting.

Applications
1) Studies on the SOS regulation in E .coli (3). For Western blotting; 1000~3000 fold dilution.
2) Construction and expression of a bait protein fused to LexA protein can be examined by Western blotting of the yeast extractsusing the antiserum.
Purified LexA protein is available from BioAcademia(#01-002)to be used as a positive control for Western blotting.
3) Immunohistochemistry (LexA fusion protein was detected in
transgenic Drosophila after fixation with 4% formalodehyde.)

Background:Mediator is a protein complex which performs a very important role both in the transcription at the basal level which does not rely on transcription control factor and in the transcription activating reaction that relies on the transcription control factor. It has the characteristic of binding to the 7 amino acid repeated structure (CTD) that exists in the C terminal of the biggest subunit of RNA polymerase II. 30 kinds of subunits have already being identified as its composing substances but it is said that multiple kinds of complexes with various subunit compositions exist in the cells. Function: AGA (Spt-Ada-Gen5 histone acetyltransferase complex) is a histone acetylase complex which has Gcn5p as a catalyst subunit and has functions overlapping with the fundamental transcription factor TFIID which has Taf1p as a catalyst subunit. However SAGA and TFIID have different allotments and each accomplishes the important role in the transcription for Housekeeping gene group and Stress Responding gene group. Also SAGA is a gigantic protein complex which is composed of Ada protein group (5 kinds) TBP related protein group (4 kinds) TAF protein group (5 kinds that also pertain to TFIID) and other protein groups (>6 kinds). It has the molecular functions such as chemical modification of histone recruitment by direct interaction of transcription regulating factor on DNA and control of transcription starting reaction by TBP. Gcn5p that shows histoneacetylase activity is one of the proteins belonging to the above-mentioned Ada protein group and it is composed of 439 amino acid recidues with molecular mass of 51 kDa in the case of budding yeast.
