Resume of DNA synthesis which was halted by various physical and chemical factors
was triggered by repair system including Rad6. Rad6 (152a.a 17.3kDa) is the
ubiqutin-conjugated enzyme E2A and complex with two Rad18 molecules ubiquitin
ligase E3 to ubiquitinate PCNA by utilizing poly-ubiquitin chain that HLTF-UBC13
synthesized and they are involved in translesion DNA synthesis (TLS) and template switch (TS). Two isoforms of Rad6 Rad6A and Rad6B differs in only 6 amino acids. They also play an important role in epigenetic transcriptional regulation by catalyzing the monoubiquitination of histone H2B at Lys-120 .

Function: DNA polymerase eta ( 713 aa 78 kDa) is specifically involved in DNA repair. Plays an important
role in translesion synthesis where the normal high fidelity DNA polymerases cannot proceed and DNA
synthesis stalls. Plays an important role in the repair of UV-induced pyrimidine dimers. Depending on the
context it inserts the correct base but causes frequent base transitions and transversions. May play a role
in hypermutation at immunoglobulin genes. Forms a Schiff base with 5-deoxyribose phosphate at abasic
sites but does not have lyase activity. Targets POLI (Pol iota) to replication foci.
Involvement in disease: Xeroderma pigmentosum variant type (XPV) [MIM:278750]: An autosomal
recessive pigmentary skin disorder characterized by solar hypersensitivity of the skin high predisposition
for developing cancers on areas exposed to sunlight and in some cases neurological abnormalities. XPV
shows normal nucleotide excision repair but an exaggerated delay in recovery of replicative DNA
synthesis. Most patients with the variant type of xeroderma pigmentosum do not develop clinical
symptoms and skin neoplasias until a later age. Clinical manifestations are limited to photo-induced
deterioration of the skin and eyes.

Function: TAFIID subunit1 (1872 aa 213 kDa) is the largest component and core scaffold of the TFIID basal transcription factor complex. Involvement in diseases: Dystonia 3 torsion X-linked (DYT3) [MIM:314250]: A X-linked dystonia-parkinsonism disorder. Dystonia is defined by the presence of sustained involuntary muscle contractions often leading to abnormal postures. DYT3 is characterized by severe progressive torsion dystonia followed by parkinsonism. It has a well-defined pathology of extensive neuronal loss and mosaic gliosis in the striatum (caudate nucleus and putamen) which appears to resemble that in Huntington disease.
