Hepatitis C virus (HCV) is a small (55-65 nm in size) enveloped positive sense single-stranded RNA virus in the family Flaviviridae and the principal cause of parenteral non-A non-B hepatitis. The virus genome consists of a single open reading frame of approximately 9.4 kb which encodes a single polyprotein of about 3010 amino acids (1 2 3). The polyprotein is processed by host cell and viral proteases into four structural proteins (core envelope1 and 2 and p7) and six non-structural proteins (NS2 3 4a 4b 5a and 5b) necessary for viral replication. HCV core protein (191 aa) is not only a component of nucleocapsid but also has multiple functions and is thought to be a pathogenic factor for hepatitis. It also participates in some cellular processes including transcriptional regulation and cellular transduction. HCV core antigen is used as diagnostic marker for HCV infection.

HIV-1 Gag p24 is a capsid protein that constitutes the core of AIDS virus HIV-1 and is produced by digestion of its precursor Gag p55 by HIV-1 protease. This protein is indispensable to the reproduction of AIDS virus and constitutes an essential element for the AIDS virus particle construction (1). p24 is used as a marker antigen for observing the patient¡Çs condition after treatment as it indicates the amount of virus in the blood.
The product is prepared by immunizing rabbit with recombinant p24 protein which was over-expressed in E. coli with a plasmid carrying the Gag p24 coding region of HIV-1 virus subtype B (2) and was highly purified by several steps of chromatography (3 4).
Using this antiserum in Western blotting the bands of 24 kD 55 kD and 41 kD corresponding respectively to HIV-p24 and its precursors p55 and p41 were observed in the extract of the AIDS virus infected cells (Fig. 1).
This product is a biotinylated IgG ([biotin]/[IgG] = 8.0) produced from the purified IgG fraction of rabbit anti-p24 serum.

Cholera toxin a main enterotoxin interacts with G proteins and increases cyclic AMP in the intestinal lining to open ion channels. As ions flow into the intestinal lumen (lining) body fluids (mostly water) flows out of the body due to osmosis leading to massive diarrhea as the fluid is expelled from the body. Cholera toxin is a complex consisting of one molecule of A subunit (27.2 kD) and 5 molecules of B subunits (11.6 kD). After secretion A subunit is proteolytically processed into A1 (22 kD) and A2 (5 kD) subunits which are held together by a disulfide bond. The toxin adsorbs to GM1 ganglioside on the surface of target cells by the B subunit and the A subunit is dissociated from the B subunit during penetration. The A subunit constitutively activates adenyl cyclase activity of ¦Á subunit of Gs (a kind of GTP-binding protein).

Schizosaccharomyces pombe Sds23 protein functions to facilitate the progression in anaphase in mitotic cell cycle. For initiating anaphase in fission yeast PP1 (protein phosphatase 1) and 20S cyclosome/APC are required. Thesds23 gene is a multicopy suppressor for mutations in PP1 and the 20S cyclosome/APC implying that the gene dosage increase can relieve the requirement for PP1 and the cyclosome/APC for the onset of anaphase. The sds23 gene encodes a 408 aa product and appears to be conserved. The sds23 gene is not essential for cell viability but in the sds23 deletion mutant the progression of anaphase and cytokinesis are retarded and cell shape is aberrant. Therefore Sds23 protein is involved in progression in anaphase as well as in cytokinesis and cell shape control. Sds23 is neither physically bound to PP1 nor a subunit of the cyclosome. It possibly regulates the PP1 and 20S cyclosome/APC in an unknown fashion.

Rad21 protein (628 aa 67.8 kDa) is a cleavable component of the cohesin complex involved in chromosome cohesion during cell cycle. The cohesin complex is required for the cohesion of sister chromatids after DNA replication. The cohesin complex apparently forms a large proteinaceous ring within which sister chromatids can be trapped. At metaphase-anaphase transition this protein is cleaved by Cut1 and dissociates from
chromatin allowing sister chromatids to segregate. Also involved in the DNA double-strand-break (DSB) repair system Hyperphosphorylated during S and G2 phases.
Proteolytic cleavage of a fraction of hyperphosphorylated form at the onset of anaphase may be essential for the proper progression of anaphase and sister chromatid separation. Belongs to the rad21 family conserved to mammals.

interacts with Nop132 to be transported to the nucleous. GTP of RagA is hydrolyzed to GDP by RCC guanine nucleotide exchange factor for RanGTPase bound to chromatin. RagA-RCC signal pathway has a crucial role in cell growth and differentiation. RagA is also well known to be involved in mTOR signaling via binding with raptor a component of mTORC1 complex in an amino acid sensitive manner.

DDX 47 gene encodes a member of the DEAD box protein family. DEAD box proteins characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD) are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation nuclear and mitochondrial splicing and ribosome and spliceosome assembly. Based on their distribution patterns some members of this family are believed to be involved in embryogenesis spermatogenesis and cellular growth and division. The protein encoded by this gene can shuttle between the nucleus and the cytoplasm and has an RNA-independent ATPase activity. Two alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.
